Constitutive XylE generator
XylE codes for the extra-diol dioxygenase called catechol 2,3-dioxygenase1. This enzyme converts catechol to 2-hydroxymuconic semialdehyde, by cleaving catechol adjacent to the two hydroxyl groups1.Because catechol is colourless and 2-HMS is a yellow compound visual assays can be used to monitor the activity of XylE. The tetR promoter (R0040) was used instead of the glucose repressible cstA promoter (K118011) so that the transformed cells could be grown in media containing glucose and XylE could still be expressed.
